One of the Growl folks provided me with a solution that solved the problem...
Try this in a terminal:
killall GrowlHelperApp
defaults write com.growl.growlhelperapp GrowlSquelchMode -bool NO
open -a GrowlHelperApp

I've done some more investigation into this and have a bit more info. I keep a separate 'vanilla user' login that I can test issues like this from. If I log in as this user, growl works great! FIXED: My growl has stopped working
I've been running growl for several years on my MBP (10.5.6). Recently it stopped working. I am not running any Logitech software, but I am running Safari 4. The apps that use growl (Unplugged for example) never show up now when growl is running, but notify me ok (thru a pop up dialog) when it isn't...
