Have you tried relaunching iTunes so that Growl may pick it up?

Applications add themselves. If an application doesn't add itself, there's a good chance that it doesn't support Growl in the first place (especially if it's not on our list). iTunes, for one, does not support Growl.

To get Growl notifications of iTunes track changes, you need an application that can translate iTunes' own track-change notifications (which are not visible to you) into Growl notifications. One such app is Quicksilver. Another is GrowlTunes, which is on the Growl disk image.
